神经酰胺代谢及凋亡信号调节

摘 要:鞘磷脂途径是一普遍存在的信号系统。神经酰胺在该途径中起第二信使分子作用。一系列合成酶或分解酶参与神经酰胺在细胞内的代谢。神经酰胺激活大量应激相关酶,如神经酰胺激活的蛋白激酶、激酶抑制因子、Jun氨基末端激酶、蛋白激酶Cζ、蛋白磷酸酶1和蛋白磷酸酶2A等,介导细胞凋亡,在应激反应诱导的疾病或肿瘤治疗中起着重要作用。本文综述近年来有关神经酰胺及其代谢物在应激反应级联以及细胞凋亡中的研究进展。

Regulation of metabolism and apoptotic signal by ceramide

Abstract: The sphingomyelin pathway is a ubiquitous signaling system, in which ceramide acts as a second messenger. Some enzymes are involved in generation or abolishment of ceramide. In several cell systems ceramide links to the stress-activated protein kinase, c-Jun N-terminal kinase, kinase suppressor of Ras, PKCζ、protein phosphatase 1, protein phosphatase 2A to signal apoptosis. The engagement of the sphingomyelin pathway in signaling apoptosis is tightly regulated to some stress related diseases and tumors. This review describes the known role of ceramide and the metabolite in stress response cascade and apoptosis.
