Necdin-Wnt及STRA6-JAK2-STAT5信号通路与肝星状细胞活化的关系研究

摘 要:肝纤维化的发生与多种信号通路密切相关,有研究证实Wnt 信号通路促进肝星状细胞(hepatic stellate cell, HSC) 活化。Necdin 作为经典Wnt 信号通路的上游信号分子,其与Wnt10b 启动子结合,激活Wnt 信号通路, 通过抑制PPARγ, 促进HSC 活化。STRA6-JAK2-STAT5 信号通路可以通过正向调节PPARγ 维持HSC 的静止。因此,研究Necdin-Wnt 和STRA6-JAK2-STAT5 信号通路与HSC 活化之间的关系,有助于为肝纤维化临床治疗提供一些新的思考。

Study in Necdin-Wnt and STRA6-JAK2-STAT5 signaling pathway and HSC activation

Abstract: Hepatic fibrosis has a close relationship with multiple signaling pathways. Previous studies have established that there is a link between HSC activation and Wnt signaling pathway. Necdin, the upstream molecule of canonical Wnt signaling pathway, binds the Wnt10b promoter in activated HSCs via the inhibition of PPARγ. The STRA6-JAK2-STAT5 signaling pathway may maintain HSC in a static state via up-regulation of PPARγ. This review focuses on the action mechanism of Necdin-Wnt as well as the relationship between STRA6-JAK2-STAT5 signaling pathway and HSC activation.
