低氧与肿瘤

摘 要: 肿瘤细胞的快速生长使得肿瘤内部缺氧,导致低氧诱导因子HIF-α 的积累。HIF-α 可以引起多种基因的转录,使得肿瘤细胞能够耐受低氧环境。综述了肿瘤细胞感受和传导低氧信号的机制,并从肿瘤细胞增殖、凋亡、代谢、迁移、浸润、血管生成以及炎症和药物耐受等多方面论述了HIF-α 在肿瘤发生发展中所发挥的重要作用。以HIF-α 为靶点是治疗肿瘤的重要手段和方法。

Hypoxia and tumors

Abstract: Hypoxia occurs in solid tumors as a result of an inadequate supply of oxygen, due to rapid cell proliferation and inefficient vascular supply. Hypoxia induces expression of hypoxia-inducible factor α (HIF-α) that activates transcription of many genes, which may help tumors adapting to the environment of low oxygen. HIF-α plays a critical role in tumor cell proliferation, survival, metabolism, metastasis and angiogenesis. It is also related to inflammation and drug resistance. Therefore, HIF-α is becoming an important target for cancer therapy.
