MFS超家族转运蛋白结构与分子机制的研究

摘 要:主要协助转运蛋白超家族(major facilitator superfamily,MFS) 是一个主要的次级膜转运蛋白超家族。MFS 超家族蛋白转运底物的多样性使得它们在细胞物质交换和能量代谢过程中起着重要作用。从2003 年第一个高分辨率的LacY 蛋白三维结构的解析到现在,已经有5 个细菌MFS 超家族的蛋白结构被解析出来,结合大量的生化研究结果,使得对其转运的分子机制有了更为深入的理解。将对MFS 超家族蛋白的三维结构和转运机理进行阐述。

Molecular mechanisms of the major facilitator superfamily transporters

Abstract: The major facilitator superfamily (MFS) constitutes one of the largest secondary active membrane transporter families found on the earth. The diversity of its substrates endows the MFS superfamily with significant role in the interchange of solutes and energy metabolism. Since the first high resolution structure of LacY determined in 2003, crystal structure of five members of the MFS superfamily have been obtained. Combined with the biochemical and biophysical investigations, we now have a better understanding of their transport mechanisms. The review summarized the advancement in the structural and mechanical understanding of the MFS superfamily transporters.
