硫酸乙酰肝素蛋白聚糖的功能机制研究进展

摘要:硫酸乙酰肝素蛋白聚糖是由核心蛋白和与之相连的硫酸乙酰肝素糖链组成,广泛分布于细胞膜与细胞外基质中。其中多配体蛋白聚糖(syndecan)和糖基磷脂酰肌醇锚定蛋白聚糖(glypican)存在与细胞膜上,而串珠蛋白聚糖(perlecan)和组合蛋白聚糖(agrin)表达在细胞外基质中。该类蛋白在生理与病理历程中,如发育、伤口愈合、肿瘤发生发展、感染、免疫应答等过程中担任重要作用,这些功能是其核心蛋白和糖链共同作用的结果。概述硫酸乙酰肝素蛋白聚糖的功能及其机制研究进展,同时强调其在作为药物靶标和临床诊断研究中的应用。

Progress in function and mechanism study of heparan sulfate proteoglycan

Abstract: Heparan sulfate proteoglycan (HSPG) is a glycoconjugate composed of core protein to which heparan sulfate chains are attached. They are widely distributed on the cell membrane and extracellular matrix. Syndecans and glypicans are the main heparan sulfate proteoglycan located on the cell membrane, while perlecan and agrin are the most investigated heparan sulfate proteoglycan in the extracellular matrix. They play an important role during physiological and pathological conditions including development, wound healing, cancer development, infection, immune response. Their function can be attributed to both the core protein and the attached heparan sulfate chains. In this review, we would like to summarize the recent progress of the heparan sulfate proteoglycan research. In the meantime, we will also highlight the potential of heparan sulfate proteoglycan as target for drug discovery & development and marker for clinical diagnostics.
