转录因子Pdx1在胰腺发育及糖尿病中的研究进展

摘 要:Pdx1 (pancreatic and duodenal homeobox 1) 是一种同源框转录因子,在胰腺发育及β 细胞功能维持中起重要调控作用。研究发现,Pdx1 基因纯合缺失会导致胰腺发育不全;在β 细胞成熟与分化过程中,Pdx1 通过协同其他因子或以顺式调控的方式参与调节;此外,Pdx1 可以直接与胰岛素启动子结合,调节胰岛素合成与分泌。近来针对Pdx1 的研究表明,它在β 细胞重塑、基因编辑及药物开发等方面具有重要作用。现就Pdx1 在胰腺发育中的作用机制及其在糖尿病治疗中的进展进行综述。
 
The role of Pdx1 in pancreas development and diabetes

Abstract: Pdx1 (pancreatic and duodenal homeobox 1) is one of the homeodomain transcription factors, and it is essential in pancreas development and islet β-cell function. It has been reported that homozygous deficiency of Pdx1 in mice leads to pancreatic agenesis. Pdx1 can also regulate β-cell maturation and differentiation by cooperating with other factors or cis-acting. In addition, Pdx1 increases insulin secretion through directly binding to and activating insulin promoter. In recent studies, Pdx1 is proved to play an important role in generation of β-like cells, gene editing and drug development. Therefore, this article mainly reviews the role and molecular mechanisms of Pdx1 in pancreas development and diabetes.
