PURA基因的结构与功能研究进展

摘 要:富含嘌呤元件结合蛋白α (purine-rich element binding protein alpha, PURα) 基因编码富含嘌呤的DNA 和RNA 结合蛋白。PURα 在进化过程中高度保守。PURα 蛋白既能直接或间接与DNA 结合发挥转录因子的作用,促进或抑制下游基因的转录,也能通过与mRNA 结合并影响其转运和翻译。PURα 的结构变异与神经系统发育和HIV 感染等多种疾病相关。近年发现PURα 的结构和功能变异也与肿瘤发生发展相关。PURα 通过影响DNA 损伤修复而影响肿瘤细胞对化疗药物的敏感性。作为转录因子,PURα 影响肿瘤相关基因的表达。PURA 基因突变和PURα 蛋白结构变异与肿瘤发生及肿瘤耐药性相关。
 
Advances in the structure and function of PURA gene

Abstract: Purine-rich element binding protein alpha (PURα) is a ubiquitous multifunctional protein that is strongly conserved throughout evolution. PURα binds directly or indirectly to DNA and functions as transcription factor; in addition, PURα binds to mRNA to influence its transport and translation. The structural variation of PURα is involved in a variety of diseases including the nervous system development and HIV infection. Recently research found that the structural and functional variation of PURα is related to the development of tumor. PURα can affect cancer cell DNA damage repair system and influence the sensitivity to chemotherapy drugs. As a transcription factor, PURα influence tumor associated gene expression. PURA gene mutation and variation of PURα can affect tumorigenesis and tumor resistance to drugs.
